Standard curve and amplification plots obtained for single qPCR assays. (a) Reactions containing single type-specific primer pairs and probes were performed
on a serial dilution of purified plasmids to generate standard curves that show a
linear relationship between copy number and Ct value in a range from 2 × 101 to 2 × 106 copies/reaction. (b) Amplification plots for each excitation/emission spectrum are
shown for each reaction containing a single type-specific primer pair and probe. Arrows
indicate the amplification plots for reactions containing a mixture of each HPV type-specific
L1 plasmid at 2 × 104 copies/reaction relative to the amplification plot corresponding to 2 × 104 copies for each type-specific HPV L1 standard. Arrows in the HPV 16 and 18 type-specific
L1 reactions also indicate the amplification plots of CaSki (HPV16), SiHA (HPV16)
and HeLa (HPV18) genomic DNA, respectively.
